Detection of Metabolite–Protein Interactions in Complex Biological Samples by High-Resolution Relaxometry: Toward Interactomics by NMR

چکیده

Metabolomics, the systematic investigation of metabolites in biological fluids, cells or tissues, reveals essential information about metabolism and diseases. Metabolites have functional roles myriads processes, as substrates products enzymatic reactions, but also cofactors regulators large numbers biochemical mechanisms. These functions involve interactions with macromolecules. Yet, methods to systematically investigate these are hitherto still scarce. In particular, there is a need for techniques suited identify characterize weak metabolite-macromolecule directly complex media such fluids. Here, we introduce method between macromolecules Our approach based on high-resolution NMR relaxometry does not require any invasive procedure separation step. We show that can detect small molecules human blood serum quantify size complex. work opens way investigations metabolite-(or other molecules) protein fluids interactomics pharmaceutical applications.

hydrochlorothiazide detection in urine samples by hplc-dad and experimental design dispersive l-l microextraction

hydrochlorothiazide (hct) is a diuretic agent which is shown to be effective in the treatment of hypertension. literature reports have demonstrated that urinary excretion data may be used to assess the bioavailability of various formulations containing this thiazide. also hct consumption by the athletes is one of the drugs which should be regulated by world anti-doping agency (wada), because of...

extremal region detection guided by maxima of gradient magnitude

a problem of computer vision applications is to detect regions of interest under dif- ferent imaging conditions. the state-of-the-art maximally stable extremal regions (mser) detects affine covariant regions by applying all possible thresholds on the input image, and through three main steps including: 1) making a component tree of extremal regions’ evolution (enumeration), 2) obtaining region ...

A high-resolution 2D J-resolved NMR detection technique for metabolite analyses of biological samples

NMR spectroscopy is a commonly used technique for metabolite analyses. Due to the observed macroscopic magnetic susceptibility in biological tissues, current NMR acquisitions in measurements of biological tissues are generally performed on tissue extracts using liquid NMR or on tissues using magic-angle spinning techniques. In this study, we propose an NMR method to achieve high-resolution J-re...

Nanosecond Time Scale Motions in Proteins Revealed by High-Resolution NMR Relaxometry

Understanding the molecular determinants underlying protein function requires the characterization of both structure and dynamics at atomic resolution. Nuclear relaxation rates allow a precise characterization of protein dynamics at the Larmor frequencies of spins. This usually limits the sampling of motions to a narrow range of frequencies corresponding to high magnetic fields. At lower fields...

Detection of Microsatellite Instability by High-Resolution Melting Analysis in Colorectal Cancer

Background: Colorectal cancer (CRC) is the third most common cancer worldwide. microsatellite instability (MSI) is a molecular marker of a deficient mismatch repair system and happens in almost 15% of CRCs. Because of a wide frequency of MSI+ CRC in Iran compared to other parts of the world, the importance of screening for this type of cancer is highlighted. Methods: The most common MSI detecti...
